I examine the mechanisms of transmission at the hippocampal mossy fiber synapse, a key synapse in the trisynaptic circuit of the hippocampus. This synapse has the main advantage that the presynaptic terminals are large, allowing direct presynaptic recording with biophysical rigor. The goal of the project is to determine the conditions under which a mossy fiber terminal initiates spikes in postsynaptic target cells, a process referred to as detonation. To address this important question, I make paired recordings between mossy fiber terminals and synaptically connected CA3 pyramidal neurons. By stimulating presynaptic terminals in the cell-attached configuration, presynaptic terminals can be stimulated highly selectively and in a noninvasive manner.
